Paradigm and Framework of WUS-CLV Feedback Loop in Stem Cell Niche for SAM Maintenance and Cell Identity Transition

چکیده

Shoot apical meristem (SAM) consists of stem cells that act as a reservoir for the aerial growth. It plays an important role in differential architectural development plants. SAM actively performs parallel functions by maintaining pluripotent and continuous organogenesis throughout plant’s life cycle. Molecular mechanisms regulating signaling networks this dual function have been progressively understood. In SAM, feedback loop WUSCHEL (WUS)-CLAVATA (CLV) has found to be key regulator stabilizing cell proliferation differentiation. general, WUS migrates into central zone (CZ) from organizing center (OC) activates expression CLV3 binding promoter elements. acts ligand interact with CLV1, leucine rich repeats (LRR) receptor-like kinase (RLK) LRR protein CLV2, coryne (CRN) (CLV2/CRN) restrict transcription OC. Cytokinin signaling as a positional cue for patterning the apical-basal axis of the growing Arabidopsis shoot meristem.

The transcription factor WUSCHEL (WUS) acts from a well-defined domain within the Arabidopsis thaliana shoot apical meristem (SAM) to maintain a stem cell niche. A negative-feedback loop involving the CLAVATA (CLV) signaling pathway regulates the number of WUS-expressing cells and provides the current paradigm for the homeostatic maintenance of stem cell numbers. Despite the continual turnover ...

Stem cell maintenance in a different niche

To overcome the difficulty of controlling stem cell fate and function in applications to regenerative medicine, a number of alternative approaches have been made. Recent reports demonstrate that a non-cellular niche modulating the biophysical microenvironment with chemical factors can support stem cell self-renewal. In our previous studies, early establishment was executed to optimize biophysic...

The Stem Cell Population of Arabidopsis Shoot Meristems Is Maintained by a Regulatory Loop between the CLAVATA and WUSCHEL Genes

The higher-plant shoot meristem is a dynamic structure whose maintenance depends on the coordination of two antagonistic processes, organ initiation and self-renewal of the stem cell population. In Arabidopsis shoot and floral meristems, the WUSCHEL (WUS) gene is required for stem cell identity, whereas the CLAVATA1, 2, and 3 (CLV) genes promote organ initiation. Our analysis of the interaction...

A Bistable Switch Mechanism for Stem Cell Domain Nucleation in the Shoot Apical Meristem

In plants, the stem cells residing in shoot apical meristems (SAM) give rise to above-ground tissues (Aichinger et al., 2012). Hence, the maintenance of stem cell niches is of central importance to a plant’s continued growth and development (Fletcher and Meyerowitz, 2000; Gordon et al., 2009). For the flowering plantArabidopsis thaliana, the genetic determinants of stem cell growth, division, a...
